The Hotel du Roy of Trois-Rivières is located between the Quebec City and the city of Montreal. 

From Montreal:

Highway 40 East, exit 196 south. Highway 55 south. Exit 181 ( Gene-H.-Kruger blvd / Notre-Dame Street). At the traffic light, turn left on Gene-H.-Kruger Blvd and continue straight ahead during about 5 minutes.

From Québec:

Highway 40 West. Exit 198 ( des Récollets Blvd). Turn left on Des Récollets blvd. Turn left again on Gene-H.-Kruger Blvd.

From Drummondville (highway 20):

Follow the instruction to go to Trois-Rivières (highway 55 north). You will cross the bridge and it will be the exit right after: 181 ( Gene-H.-Kruger blvd / Notre-Dame Street).  At the traffic light, turn left on Gene-H.-Kruger Blvd and continue straight ahead during about 5 minutes.
